Doktorarbeit / Dissertation aus dem Jahr 2003 im Fachbereich Medizin - Neoplasmen, Onkologie, Note: magna cum laude, Johann Wolfgang Goethe-Universität Frankfurt am Main (Uniklinik, Zentrum der Inneren Medizin), Sprache: Deutsch, Abstract: Obwohl es bereits seit dem Altertum gewisse Kenntnisse über die tumorreduzierende Wirkung von Pflanzen und Mineralien gab, konnte erstmals der Breslauer Neurologe Heinrich Lissauer im Jahr 1865 in einer wissenschaftlichen Arbeit über positive Effekte einer Chemotherapie bei chronischer myeloischer Leukämie (CML) berichten.1 Zwei Patienten mit CML zeigten Zeichen einer Teilremission, nachdem sie mit Kaliumarsenit (Fowler'sche Lösung) behandelt worden waren, ein Effekt, der 1931 durch C. E. Forkner wiederentdeckt wurde.2 1892 berichtete William B. Coley über antineoplastische Effekte von Bakteriengiften (ein Bakterientoxin-Cocktail namens "Coley's Toxin") auf verschiedene Tumoren, entdeckt als eine Nebenwirkung von postoperativen Wundinfekten bei Sarkompatienten. Mit der Entwicklung von Salvarsan (1909) gilt Paul Ehrlich als Begründer der modernen Chemotherapie, obgleich diese Substanz weniger antineoplastische als vielmehr antibakterielle Wirkung hatte.3,4 Einen mehr biologischen Weg wählte sein Freund und zugleich Konkurrent Emil von Behring, der bereits zwanzig Jahre zuvor mit der Serumtherapie den Grundstein der heutigen Immuntherapie gelegt hatte.5 Die Entwicklung von Senfgas (Lost) durch Alfred Gilman im 1. Weltkrieg richtete das Augenmerk auf eine Substanz, die neben der Wirkung als Reiz- und Kampfgas auch starke myelosuppressive Effekte aufwies (u.a. reduzierte Lymphozytenzahl). Um die Giftigkeit zu reduzieren, wurden Schwefel- und später Stickstofflostverbindungen hergestellt, die allerdings immer noch sehr toxisch waren.
