Le roman raconte le périple de Michel Strogoff, courrier du tsar de Russie de Moscou à Irkoutsk, capitale de la Sibérie orientale. Sa mission est d'avertir le frère du tsar, resté sans nouvelles de Moscou, de l'arrivée des hordes tatares menées par le traître Ivan Ogareff pour envahir la Sibérie. Sur cette route pleine d'obstacles, il croise la belle Nadia, ainsi que les journalistes européens Harry Blount et Alcide Jolivet. Il n'a que cinq semaines pour aller de Moscou à Irkoutsk. Les courriers du tsar mettent à peine dix-huit jours pour parcourir cette distance. Michel Strogoff met trois mois, à cause de toutes les épreuves qu'il doit surmonter. Par exemple, il se fait brûler les yeux et devient aveugle... Nadia, sa fidèle amie, le suivra tout au long de son aventure. Ils se font passer pour frère et soeur.
