Sickle cell anemia is a hereditary blood disorder characterized by the presence of abnormal hemoglobin, known as hemoglobin S, which leads to the formation of sickle-shaped red blood cells. This genetic condition causes a range of complications, including pain crises, anemia and organ damage. The disease primarily affects individuals of African, Mediterranean, Middle Eastern and Indian descent.
Sickle cell anemia results from specific mutation in the HBB gene, leading to the production of hemoglobin S. The altered hemoglobin causes red blood cells to become rigid and take on a characteristic sickle shape. The deformed cells can clump together, leading to blockages in blood vessels and causing vaso-occlusive crises, one of the hallmark features of the disease.
Understanding the genetic basis of sickle cell anemia is essential for grasping the mechanisms that underlie the disorder. The HBB gene is located on chromosome 11. A single nucleotide substitution results in the replacement of glutamic acid with valine in the hemoglobin protein.
